The protocol at a glance
There is no approved nasal dose. Research trials commonly use a single 24 IU nasal dose in adults. The largest children's autism trial targeted 48 IU a day for 24 weeks and found no benefit over placebo.
| Study or label | Amount |
|---|---|
| Approved nasal dose | None found |
| Common single nasal research dose, adults (68-man trial, 2023) | 24 IU |
| SOARS-B autism trial, children 3 to 17, 24 weeks | Target 48 IU a day, nasal |
| Pitocin label (injection, hospital use) | Given by drip by staff; not a nasal or home product |

Community reports commonly describe 20-40 IU, often nasal.

Mixing
No mixing table here: this one is used as a capsule, nasal spray or skin product, or has no human amount to calculate from. The general mixing calculator works for any vial.
What the trials found
SOARS-B: 290 children and teens with autism, 24 weeks, nasal oxytocin vs placebo, target 48 IU a day. Measured: change in a social withdrawal score (lower is better):
Sikich L et al. N Engl J Med 2021 (PMID 34644471). Other secondary outcomes generally did not differ, and side effects were similar to placebo.
Side effects
- In the autism trial, the number and severity of side effects were similar to placebo.
- Injected oxytocin (Pitocin) in labor can cause uterine overstimulation or rupture at excess doses, irregular heartbeat, and severe water intoxication with fits if infused over a long time. This is a hospital drug and not the same use as a nasal spray.
- Nasal oxytocin did not change eye gaze or bonding in a 2023 trial of 68 men at 24 IU.
How it works
Oxytocin receptors
It is a natural hormone. In labor it makes the womb contract; the label uses it to start labor and control bleeding after birth.
Nasal delivery is uncertain
How much of a nasal spray reaches the brain is debated, and delivery varies between people and devices (Guastella 2013).
