The protocol at a glance
There is no tested schedule for this blend in people. The closest human data are separate: a long-lasting CJC-1295 (with DAC) given weekly or every other week to healthy adults, and ipamorelin given into a vein.
| What was tested | Amount and schedule |
|---|---|
| Human dose of this blend | Not established |
| CJC-1295 WITH DAC (a different, long-acting form), healthy adults | 30 or 60 mcg per kg under the skin, single doses or 2-3 doses a week or two weeks apart (28- and 49-day trials) |
| Ipamorelin, healthy men | One 15-minute drip into a vein, from 4.21 up to 140.45 nmol per kg |
| Ipamorelin, after bowel surgery | 0.03 mg per kg into a vein, twice a day, up to 7 days |
Sources: Teichman 2006 (CJC-1295 with DAC), Gobburu 1999 and Beck 2014 / NCT00672074 (ipamorelin). CJC-1295 without DAC lacks the long-acting part, so these amounts should not be copied across.
Community reports commonly describe 100-250 mcg of each peptide under the skin before bed, daily, for 8-12 weeks. These are community reports, not trial schedules.
What people say they do, collected from community discussions. Not tested in a trial; shown so you can compare it with the evidence above.
Mixing
No mixing table here: this one is used as a capsule, nasal spray or skin product, or has no human amount to calculate from. The general mixing calculator works for any vial.
What the trials found
Healthy adults, placebo-controlled trials of 28 and 49 days using CJC-1295 WITH DAC (Teichman 2006). This is not the no-DAC form. Measured growth hormone (GH) and IGF-1 in blood:
Teichman SL et al. J Clin Endocrinol Metab 2006 (PMID 16352683); Gobburu JV et al. Pharm Res 1999 (PMID 10496658). In the one ipamorelin trial that looked at a health outcome (bowel recovery after surgery, 114 patients, NCT00672074), it did not beat placebo: median 25.3 vs 32.6 hours to first meal, p = 0.15 (Beck 2014, PMID 25331030).
Side effects
- CJC-1295 with DAC: no serious adverse reactions were reported in the healthy-adult trials, and it was called relatively well tolerated, especially at 30 or 60 mcg per kg.
- Ipamorelin 0.03 mg per kg into a vein twice a day for up to 7 days was well tolerated after bowel surgery. Side effects were as common as with placebo (87.5% vs 94.8%).
- A 2006 phase 2 study of CJC-1295 in people with HIV (NCT00267527) was stopped early and is listed as terminated. News reports at the time linked the stop to a participant's death. I could not read the company's own report, so the cause is not confirmed here.
- Long-term safety of either peptide, or of the blend, is unknown.
How it works
Two different signals
CJC-1295 is a growth-hormone-releasing-hormone (GHRH) analog that tells the pituitary gland to release GH. Ipamorelin works through a ghrelin-type receptor and also triggers GH release.
GH pulses stay normal (CJC-1295 with DAC)
In healthy men, one dose of the long-acting form kept normal GH pulses but raised the low points between them (Ionescu 2006).
